Replacement proteins
The first generation of biotherapeutics were largely natural proteins used to replace missing proteins or boost physiological function, such as human growth hormone and erythropoietin; yet the vast majority of natural proteins have attributes that limit their use or make them suboptimal as therapeutics.
Traditional protein engineering
Traditional protein engineering includes very successful manipulation of antibodies, as well as simple fusions such as Fc fusions. The breadth of proteins whose structures could be confidently manipulated has been confined to just a few structural classes.

In the last 10 years, there has been an exponential increase in the understanding of biophysical characteristics and structure-function relationships of proteins and antibodies. At Eleven, we create first-of-a-kind protein designs by employing a holistic view to the creation of next generation protein therapeutics using an integrated understanding of how proteins interact with their targets at the molecular level to improve the impact of the drug on disease processes. Starting with clinically-validated pathways and well-established biology and biochemistry, Eleven expands beyond simple protein engineering by using tools and technologies from our AMP-Rx product engine that enable us to rationally design novel protein structures with ideal pharmacologic characteristics to achieve an optimal product profile to target a disease. This approach not only overcomes traditional protein engineering challenges and limitations, but has inherent speed and productivity advantages by leveraging multiple tools and rapid testing to reduce the time required from concept to preclinical validation.
